Hedyosmum goudotianum Solms extract controls inflammation by suppressing NF-¥êB and STAT3 pathways in LPS-activated macrophages

Abstract
Although inflammatory reactions are a mechanism to protect against microbial invasion in the body, however, in excessive cases, they develop into chronic inflammatory diseases. Macrophages stimulated by the lipopolysaccharide (LPS), one of the outer membrane components of Gram-negative bacteria, induce an inflammatory response by activating various transcription factors and expressing inflammatory mediators. Therefore, it is important to reduce the expression of pro-inflammatory cytokines and inflammatory mediators to suppress the excessive inflammatory reaction. In this study, we report the immunosuppressive effect of Hedyosmum goudotianum Solms (H. goudotianum) extract in response to LPS using RAW264.7 macrophages and suggest the signaling mechanism. Hedyosmum goudotianum extract effectively inhibited LPS-induced expression of inflammatory mediators such as inducible nitric oxide synthase (iNOS), tumor necrosis factor (TNF-¥á), interleukin-1¥â (IL-1¥â), and interleukin-6 (IL-6). Moreover, H. goudotianum extract suppressed the activation of nuclear factor-¥êB (NF-¥êB) and signal transducer and activator of transcription 3 (STAT3), which are known to be important transcription factors in the expression of pro-inflammatory cytokines. Therefore, we suggest that H. goudotianum extract has a potential value to suppress an excessive inflammation caused by the microbial invasion.
